Chemistry 321L ManualPage 32 CaffeineCaffeine UV SpectrumHigh Performance Liquid ChromatographyI. IntroductionMany beverages such as soft drinks, coffee and tea contain the mild stimulant caffeine(C8H10N4O2). The caffeine content varies widely from about 100 g/mL (100 ppm) in sodas toover 1000 g/mL in certain types of coffee. In this experiment the caffeine content of a dilutedsoft drink will be determined using high Performance Liquid Chromatography (HPLC).The chemical structure of caffeine is shown provides a sensitive method for the detectionand measurement of caffeine. The UV absorption spectrum (seefigure below) of caffeine exhibits a pair of absorption bandspeaking at 205 nm and 273 nm with a characteristic absorptionshoulder between them.
