METHANOL LEAFEXTRACTSSiti Nur Sarah, Kamaruzaman Sijam and Dzolkhifli OmarDepartment of Plant Protection, Faculty of Agriculture, Universiti Putra Malaysia, 43400 UPM Serdang, Selangor, methanol leaf extract ofPsidium guajavaL. was prepared and investigated for the existence of chemical constituentsusing thin layer chromatography (TLC). It was concluded thatP. guajavamethanol leaf extracts possess numerous ofchemical constituents represented by different retention factor (Rf) : PSIDIUM guajavaL., thin layer chromatography (TLC), retention factor (Rf).INTRODUCTIONOver decades, PSIDIUM guajavaL. were known as a treewhich served the food cropand medicinal plant in tropicaland subtropical countries around the world. All of theguava tree parts are useful to human as it serves manyuseful properties instead of food and medicine.

Guava hasbeen used widely in pharmacological, traditional, clinicaland analysis of chemical constituents. These trees give agood yield of useful chemical constituents which possessnumerous of biological activities belong to alkaloid, flavonoid, triterpene, saponin, terpenoid, carotenoid, andtannin. Most of these active chemical constituents presentswithin guava fruits and leaves. Due to the presence ofthese active chemical constituents had turned guava treeinto a tree that were active with pharmacological activitiessuch as anti-allergy, cytotoxic, antimicrobial, hepatoprotection, antioxidant and antimicrobial (Guti rrezet al., 2008).Previous studies performed onP. guajavaleaves haveresulted in the presence of essential oil such as cineol, tannins and triterpenes. There were also three differenttypes of flavonoids as quercetin, avicularin andguaijaverin isolated from the leaves (Khadem andMohammed, 1959).

The use of thin layer chromatography(TLC) plate for extracts PROFILING , will indicate thepresence of numerous chemical constituents representedby different retention factor (Rf) values, without revealingthe exact names for each compounds. The objective of thisstudy was to examine the presence of chemicalconstituents withinP. guajavamethanol leaf AND METHODSP lant materialThe fresh leaves ofPsidium guajavawere collected fromuniversity agricultural park, Universiti Putra Malaysia,Serdang, Selangor. The fresh leaves were clean underrunning tap water and leaves to dry for two weeks at roomtemperature. Dry leaves were crushed with coffee grinderinto fine powder and stored in an air tight container forfurther extractionTwo hundred grams ofP. guajavaleaves powder wassoaked into 2 L of absolute methanol and left at roomtemperature for two days with occasionalshaking.

Theextracts were filtered through Whatman No. 1 filter paperinto a clean flask and lyophilized to dryness using rotaryevaporator. The dry extracts obtained than were kept infreezer at 4 C until further application and developmentAvolume of 10 l of methanol extract was applied on 2 x5 cm silica gel 60 F254plate of mm thickness. The platewas then developed in a solvent system hexane : ethylacetate (7 : 3) and visualized under normal daylight. TheRfvalues were calculated for each bands revealed underdaylight. The formula used for Rfvalues calculation wasas AND DISCUSSIONThe presence of number of chemical constituents wasproved through TLC PROFILING . Different Rfvaluesrepresent different chemical constituents presents withinmethanol leaf extract ofPsidium was done as the compound exist on TLCplate surface were naturally colored and easily can bedetected and calculated for Rfvalues.

From the experimentconducted, there were six visible spots were detected whenvisualized under daylight as shown in Figure 1. The Rfvalues for each spot were as depicted in Table travelled by the substanceDistance travelled by the solventTLC PROFILING ofPsidium leaf extracts553 FIGURE 1:TLC spot visualization ofPsidium guajavamethanol leaf extract under daylightTABLE 1:The retention factor (Rf) values ofPsidiumguajavamethanol leaf extract visualized under different Rfvalues obtained from TLC PROFILING ofPsidium guajavamethanol leaf extracts indicated thepresence of numerous of active chemical constituentswithin the rrez, , Mitchell, S. and Solis, (2008) PSIDIUM guajava:A review of its traditional uses, phytochemistry and (1): , and Mohammed, (1959)Constituents of the leavesofPsidium Chemical.
