KIN 505 Research Laboratory Techniques in exercise physiology Rick Sharp 250 Forker Bldg 294-8650 Required Text: KIN exercise physiology Lab Manual (supplied on Blackboard) plus handouts I will post on Blackboard usually these are a variation of what s in the lab manual. This course is designed to introduce you to some of the analytical methods for the study of the physiological and biochemical responses to exercise . Tentative Course Outline We will meet as a group on Mondays for the main purpose of introducing the lab experiment that you will conduct during an arranged day and time convenient for your lab group (some lab experiments will take multiple days to complete hence will span a couple of weeks).

In addition to these scheduled times, you may also arrange to use the lab during other times for purposes of gaining more practice with a method or to repeat an experiment that went haywire . Your initiative in doing whatever it takes to perfect your lab Techniques will pay great dividends in your graduate and professional life. More immediately, initiative is noticed and affects your grade in the class. This is more of a doing class and can help you learn new Techniques , re-learn Techniques , or just gain a better understanding of lab methods you may read about in the literature.

Take advantage of this opportunity to learn and use these important tools of your profession. Date Topic/Activity 8/24 Introductions and Orientation (Go to EH&S Login for Trainings, Human Subjects, Lab Safety, Fire Safety) Reading: Error and Precision of Physiological and Biochemical Measurements 8/31 Error and precision of physiological measurement 9/7 Holiday 9/14 Indirect Calorimetry: Calibration check of metabolic carts I 9/21 Indirect Calorimetry: Calibration check of metabolic carts II Report: Full report 9/28 Indirect Calorimetry: Fuel Utilization I 10/5 Indirect Calorimetry: Fuel Utilization II Report.

Full Report 10/12 Body Composition (skinfold vs BodPod vs anthropometry) Report: Datasheet 10/19 Midterm Exam 10/26 Processing Blood and Blood Products (Bloodborne Pathogens, Biohazardous Materials, & Autoclave Safety) 11/2 Pipetting and Weighing/Biochemical Calculations Report: Datasheet only 11/9 Hemoglobin and Hematocrit Measurement Report: Datasheet 11/16 Blood Lactate Concentration Report: Full Report 11/23 Thanksgiving break 11/30 Blood Glucose I 12/7 Blood Glucose II Report: Full Report 12/14 Final Exam Week Departmental Outcomes and Course Objectives Learning Objectives 1) Define and evaluate principles of accurate and reproducible measurement.

2) Understand principles behind measurement of metabolic responses during exercise . 3) Demonstrate ability to troubleshoot measurement methods in various forms of exercise testing, body composition, and resting characteristics. 4) Demonstrate ability to set up and calibrate equipment to be used in testing. 5) Understand and practice safe Laboratory skills according to EH&S guidelines. 6) Define and understand basic principles of biochemical measurements in blood and other tissues. 7) Conduct both biochemical and physiological testing at rest and during exercise .

8) Communicate results of experiments in scientific writing style. 9) Demonstrate ability to work in collaboration with fellow students to conduct and analyze Laboratory -based experiments. 10) Demonstrate effective use of background literature both in setting up experiments and in evaluating results. Learning Objectives and Assignments Linked to Outcomes 1) Communication. Uses clear and effective written, oral, visual, and electronic (WOVE) communication Techniques to foster inquiry, collaboration, and engagement in physical activity and health related settings.

LO #8, #9. Assignments: Laboratory reports, midterm exam, final exam, group projects. 2) Lifelong learning, assessment, and self-reflection. Analyzes and evaluates one s own knowledge, abilities and actions relative to professional standards, seeks opportunities to grow professionally, and utlilizes self-assessment and assessment of others to foster physical, cognitive, social, and emotional well-being. LO #3, #5, #9. Assignments: Laboratory reports, group projects. 3) Content knowledge, discovery, and critical thinking. Understands fundamental concepts of physical activity and health, conducts scientific inquiry, and applies critical thinking to solve problems from personal, scholarly, and professional perspectives.

LO #1-10. Assignments: Laboratory reports, midterm exam, final exam, group projects. 4) Ethics, diversity, and social justice. Demonstrates leadership and social responsibility to improve quality of life for others and ensures equitable access for diverse groups by creating appropriate environments to initiate and maintain a physically active, healthy lifestyle. LO #2, #5, #7, #9. Assignments: Laboratory reports, midterm exam, group projects. Evaluation Except when instructed otherwise, you will submit a printed and double spaced report following each lab exercise .

There are two main purposes of these reports: 1) to give you practice in writing in the style used in scientific papers ( , journal articles), and 2) to condition your brains to begin thinking in the science pattern (identify a problem and develop a rationale/hypothesis, consider the methods and Techniques that will allow logical and appropriate examination of the problem and hypothesis, formulate a plan of analysis, present your results, and interpret your results in the context of the hypotheses and in comparison with work from other researchers). This report should take the form of a journal article containing the following items: 1.

Introduction: Background information including rationale for the study and pertinent previously published Research on this topic. Not expected to be an exhaustive review of literature, but some literature usually needs to be cited. Most of the time, using about 2-4 literature citations is sufficient. 2. Methods: Describe what equipment was used and precisely how they were used. Also any other procedures such as how the subjects were treated, etc. Do not list - Everything belongs in narrative format (paragraphs; not bullet points). 3. Results: Present all the raw data (or means where appropriate) in either tabular or graphic form, as well as a description of the results in narrative form.
