1 Minimum Essential Medium Eagle (MEM)With Earle's salts, L-Glutamine, NEAA and Sodium pyruvateWithout Sodium bicarbonateProduct Code: AT154 Please refer disclaimer overleafProduct Description : Minimum Essential Medium (MEM) is a modification ofBasal Medium Eagle (BME). It was developed by HarryEagle to meet the specific nutritional requirements ofcertain subtypes of HeLa cells and normal mammalianfibroblasts. MEM includes higher concentration ofamino acids so as to closely approximate the proteincomposition of cultured mammalian cells. MEM can beused either with Earle's salts or Hanks salts and can alsobe additionally supplemented with Non- Essential aminoacids (NEAA). This Medium can be further modifiedby eliminating calcium to facilitate growth of cells insuspension is Minimum Essential Medium Eagle with Earle'sbalanced salts, L-glutamine, non- Essential amino acidsand sodium pyruvate. Users are advised to reviewthe literature for recommendations regarding mediumsupplementation and physiological growth requirementsspecific for different cell :Ingredientsmg/LINORGANIC SALTSC alcium chloride sulphate dihydrogen phosphate hydrochloride disodium salt red sodium salt :1.
2 Suspend in 900ml tissue culture grade waterwith constant, gentle stirring until the powder is completelydissolved. Do not heat the Add of sodium bicarbonate powder (TC230) of bicarbonate solution (TCL013) for 1litre of Medium and stir until Adjust the pH to - pH units below the desired pHusing 1N HCl or 1N NaOH since the pH tends to rise Make up the final volume to 1000ml with tissue culturegrade Sterilize the Medium immediately by filtering through asterile membrane filter with a porosity of micron or less, using positive pressure rather than vacuum to minimize theloss of carbon :User must ensure suitability of the product(s) in their application prior to use. 4 All the powdered media and prepared liquid culture mediashould be stored at 2-8 C. Use before the expiry date. In spiteof above recommended storage condition, certain powderedmedium may show some signs of deterioration /degradationin certain instances. This can be indicated by change incolour, change in appearance and presence of particulatematter and haziness after Preparation of concentrated Medium is not recommendedsince free base amino acids and salt complexes having lowsolubility may precipitate in concentrated pH and sodium bicarbonate concentration of the preparedmedium are critical factors affecting cell growth. This isalso influenced by amount of Medium and volume of culturevessel used (surface to volume ratio). For example, in largebottles, such as Roux bottles pH tends to rise perceptibly assignificant volume of carbon dioxide is released. Therefore, optimal conditions of pH sodium bicarbonate concentration, surface to volume ratio must be determined for each cell recommend stringent monitoring of pH.
5 If needed, pHcan be adjusted by using sterile 1N HCl or 1N NaOH or bybubbling in carbon If required, supplements can be added to the Medium priorto or after filter sterilization observing sterility life of the Medium will depend on the nature ofsupplement added to the : 1 / 2011
